GLP-1 Receptor
The GLP-1 receptor (GLP-1R) is a class B G-protein coupled receptor expressed on pancreatic beta cells, central nervous system neurons, and gastrointestinal tissues. It is the target of the incretin hormone GLP-1 and synthetic agonists including semaglutide.
Activation of GLP-1R triggers cAMP/PKA signaling cascades. Published research has examined the receptor's role in glucose-dependent insulin secretion, glucagon suppression, and gastric emptying regulation.
Semaglutide, tirzepatide, and retatrutide all engage the GLP-1R, though with different selectivity profiles: semaglutide is GLP-1R-selective, tirzepatide also targets GIP-R, and retatrutide additionally engages the glucagon receptor.
