Chromatogram
A chromatogram is the graphical output of an HPLC analysis. The x-axis represents retention time (minutes) and the y-axis represents UV absorbance (mAU). Each peak corresponds to a molecular species detected in the sample.
In a peptide chromatogram, the target compound appears as the dominant (tallest) peak. Impurity peaks — deletion sequences, truncated fragments, oxidized forms — appear as smaller peaks at different retention times. The relative areas of all peaks are used to calculate purity percentage.
A high-quality chromatogram shows: a sharp, symmetrical main peak with minimal tailing, a flat baseline with low noise, and small or absent impurity peaks. A chromatogram image should be included on every COA to allow visual verification of the purity claim.
