Understanding Peptide Purity: What HPLC Analysis Really Tells You

What Is HPLC?

High-Performance Liquid Chromatography (HPLC) is the standard analytical technique for determining the purity of synthetic peptides. It separates peptide molecules based on their physical and chemical properties as they pass through a specialized column under high pressure.

The result is a chromatogram — a graph showing peaks that correspond to different molecular species in the sample. The target peptide should appear as the dominant peak, while any synthesis byproducts, truncated sequences, or degradation products appear as smaller peaks.

Reading a Purity Percentage

When we state a peptide has ≥98% purity by HPLC, this means that at least 98% of the total chromatographic area corresponds to the target peptide. The remaining ≤2% represents synthesis-related impurities — typically deletion sequences, truncated fragments, or oxidation products.

Why Independent Testing Matters

Self-reported purity from the manufacturer is a starting point, but independent third-party verification eliminates conflicts of interest. Our protocol sends each batch to an accredited laboratory that has no relationship with the synthesis facility.
