[NAD+](/peptides/anti-aging/nad-plus) (nicotinamide adenine dinucleotide) is a coenzyme present in all living cells. MW: 663.43 Da (CAS: 53-84-9). Formula: C21H27N7O14P2. It exists in oxidized (NAD+) and reduced (NADH) forms and participates in over 500 enzymatic reactions.
NAD+ serves as an essential cofactor for three major enzyme families: sirtuins (SIRT1-7, NAD+-dependent deacylases), PARPs (poly-ADP-ribose polymerases involved in DNA repair), and CD38/CD157 (ectoenzymes involved in calcium signaling).

### NR Supplementation Trial

The study reported that NR elevated circulating NAD+ metabolites by approximately 60% and was well-tolerated over the study period.
### NADPARK Study
[Brakedal et al. (2022)](https://pubmed.ncbi.nlm.nih.gov/35235774/) published results from the NADPARK study — a phase 1 randomized clinical trial of NR supplementation in participants with Parkinson's disease. The study's primary endpoint was safety and tolerability. The authors reported that NR increased cerebral NAD+ levels as measured by magnetic resonance spectroscopy.
### NMN Supplementation Trial
[Kim et al. (2023)](https://pubmed.ncbi.nlm.nih.gov/36797393/) published a randomized, double-blind, placebo-controlled trial of NMN (nicotinamide mononucleotide, another NAD+ precursor) in middle-aged adults. The study reported increases in blood NAD+ concentrations and examined walking speed and grip strength as functional endpoints.
