What Is NAD+?

Nicotinamide adenine dinucleotide (NAD+) is a coenzyme found in all living cells. It exists in two forms: NAD+ (oxidized) and NADH (reduced). The ratio between these two forms is critical for cellular energy metabolism.

NAD+ participates in over 500 enzymatic reactions and serves as an essential cofactor for three major enzyme families: sirtuins (SIRT1-7), poly-ADP-ribose polymerases (PARPs), and CD38/CD157 ectoenzymes.

The Sirtuin Connection

Sirtuins are a family of seven NAD+-dependent deacylases and ADP-ribosyltransferases. They regulate gene expression, DNA repair, metabolic pathways, and stress responses. SIRT1 and SIRT3 have been the most extensively studied in the context of aging research.

Each time a sirtuin deacetylates a substrate, it consumes one molecule of NAD+. This creates a direct link between cellular NAD+ availability and sirtuin activity.

Age-Related Decline

Published research has documented that NAD+ levels decline with age in multiple tissues. This decline has been observed in human blood samples, mouse tissues, and C. elegans models. The mechanisms of decline include increased CD38 expression, decreased NAMPT (the rate-limiting enzyme in NAD+ salvage), and increased PARP activity from accumulating DNA damage.

Research on NAD+ Supplementation

Studies using NAD+ precursors (NMN, NR) and direct NAD+ supplementation have been published in journals including Cell Metabolism, Nature Communications, and Science. These studies have examined effects on mitochondrial function, cellular respiration, and age-related biomarkers in various model systems.
